For sure you need to buy in advance as they sell out fast. Inform that station master at Kanchanaburi that you will board there and he will inform the guard on the train.

I’m already in Kanchanaburi and want to board this weekend excursion train from Kanchanaburi (not Bangkok) and go up to the Sai Yok Noi Waterfall. I like the timing better and that it stops for 20 mins at the Kwai River Bridge (far from my hotel). Is this allowed or will I be considered a “no show” in Bangkok since I’m only boarding in Kanchanaburi? And is it possible to buy train tickets at the station on the day instead of online in advance? Thanks!

This excursion train stops at the bridge for you to get out to take pictures for twenty minutes or so before getting back on to continue your journey.
